Tirzepatide vs Semaglutide: Identity and Chemistry Compared

Tirzepatide is a GLP-1/GIP dual receptor agonist. Semaglutide is a GLP-1 receptor agonist. One binds at two incretin receptors, the other at one, and that is the entire classification difference between them. Everything below is the detail behind it.

This is an identity comparison, not an effectiveness comparison. I am not ranking these two against each other on anything, because that would require talking about outcomes, and this category does not do outcomes. What it does is molecular identity: which compound is in the vial, what the number on the label refers to, and how the material is handled and tested as a reagent.

What the receptor class actually names

Receptor-target class is a naming convention. It sorts a molecule by which receptor family its sequence was built around, the same way "cyclic peptide" or "15-amino-acid fragment" sorts a molecule by shape and length. It is a statement about structure, not about consequence.

GLP-1 and GIP are both incretin receptors. Semaglutide's sequence is built around GLP-1 alone. Tirzepatide's is built to engage both GLP-1 and GIP, which is what "dual agonist" means on a catalog line. The same axis extends further: retatrutide is classified as a GLP-1/GIP/glucagon triple receptor agonist. Single, dual, triple. That is the sorting system, and it explains why these three compounds are almost always indexed next to each other.

The chemistry on the label

These are the values that settle which molecule you actually have. If you are logging a lot or comparing two suppliers, the CAS number is the one that ends arguments.

Two notes on that table.

First, I am not quoting a molecular weight figure for either compound. Published values drift depending on whether the source is describing the free base or a salt, and a formula plus a CAS number pins identity far more tightly than a rounded mass does. If you want a sense of scale in this class, retatrutide's free-base formula is C221H342N46O68 at 4731.32 g/mol, and both molecules here sit in the same neighbourhood.

Second, the formulas above are free-base formulas. Synthetic peptides of this size are commonly supplied as trifluoroacetate (TFA) salts, which changes the mass of what is in the vial without changing which peptide it is. The free-base formula stays the free-base formula; I am not restating it as a salt formula.

What the atom counts tell you

Tirzepatide carries 38 more carbons, 57 more hydrogens, 3 more nitrogens and 9 more oxygens than semaglutide. That is a substantial difference in size for two compounds that get discussed in the same breath, and it is the reason mass spectrometry separates them without ambiguity. Two peptides that differ by a single residue can be a genuine analytical problem. These two are not that case.

Where they appear in the research literature

Both compounds appear in metabolic and endocrine research, in incretin receptor pharmacology, and in analytical and peptide-chemistry method development work. Those are subject areas. Naming a field a compound has been studied in describes what researchers have looked at, not what they concluded.

Form, storage and handling
