Tirzepatide is a synthetic 39-residue single-chain peptide described in the literature as a dual incretin receptor agonist, with measurable activity at both the GIP receptor and the GLP-1 receptor. It sits in the same class as semaglutide, which acts at the GLP-1 receptor alone, and retatrutide, which adds a third target at the glucagon receptor.

Structurally the catalogue records aminoisobutyric acid (Aib) substitutions at positions 2 and 13, a C20 fatty diacid attached at Lys20 through two AEEA linkers and a gamma-glutamate spacer, and a C-terminal amide. The lipid chain is the modification the literature associates with extended circulating half-life in the models studied. CAS 2023788-19-2, formula C225H348N48O68, molar mass 4813.45 g/mol.

Tirzepatide appears in receptor-pharmacology and metabolic-research literature as the reference compound for dual GIP and GLP-1 receptor agonism, and it is frequently used as a comparator when a new incretin analogue is characterised.

Published studies have reported potency at both receptors in cell-based assays, and have examined how the balance between the two contributes to the effects seen in animal metabolic models relative to GLP-1-only agonists.
