Semaglutide

Semaglutide is a synthetic analogue of GLP-1, built on a 31-residue backbone derived from the GLP-1(7-37) sequence. The catalogue records two substitutions from the natural peptide, aminoisobutyric acid at position 8 and arginine at position 34, plus a C18 fatty diacid attached at Lys26 through two AEEA linkers and a gamma-glutamate spacer.

Each modification has a described purpose in the literature: the position-8 substitution resists cleavage by dipeptidyl peptidase-4, which degrades native GLP-1 within minutes, and the fatty diacid binds circulating albumin, which is associated with an extended half-life in the models studied. Semaglutide is the reference GLP-1 receptor agonist in incretin literature and is the standard comparator when a newer analogue is characterised.

Published studies have reported receptor potency in cell-based assays and effects in animal metabolic models, and the structure-activity work on its lipidation and DPP-4 resistance is documented in detail. Papers on tirzepatide and retatrutide routinely benchmark against it, which is why it appears in almost every comparison in this class.

A licensed prescription medicine containing Semaglutide is approved and sold in Canada under other brand names. What we supply is NOT that medicine. It is an unapproved laboratory reagent, not manufactured, released or supplied to pharmaceutical standards, and not for human or veterinary use. Read those certificates for what they are: they were issued by the laboratory named on them to our supplier for that production lot, and our batch code is the internal receipt reference we assign, so it does not appear on the PDF.
