Retatrutide vs Tirzepatide: Receptor Targets and Handling
Both compounds sit in the incretin class and both are studied as multi-receptor agonists, which is why they are constantly compared. The meaningful difference is how many receptors each is designed to engage.
Receptor targets
Tirzepatide is a dual agonist, engineered to act at two receptors:
- GLP-1 (glucagon-like peptide-1)
- GIP (glucose-dependent insulinotropic polypeptide)
Retatrutide is a triple agonist, adding a third:
- GLP-1
- GIP
- Glucagon (GCGR)
That third receptor is the entire structural distinction. Adding glucagon-receptor activity to an incretin backbone is a substantially different design problem from balancing two, because the three activities have to be tuned against one another rather than simply maximised.
Structure and synthesis
Both are synthetic analogues built on incretin backbones, modified to resist enzymatic degradation and extend circulating half-life. Both carry a fatty-acid moiety promoting albumin binding, which is the standard approach to slowing renal clearance in this class.
Both are long by synthetic peptide standards. That matters practically rather than theoretically: the longer the chain, the more coupling steps in solid-phase synthesis, and the more opportunity for deletion and truncation sequences that are chemically similar enough to be hard to separate. Handling
Neither is unusual to work with. Both arrive lyophilized, both dissolve readily in bacteriostatic or sterile water, and both follow the general pattern in the reconstitution guide: add the diluent down the vial wall, swirl rather than shake, then refrigerate the solution and keep it dark.
Lyophilized vials are held frozen for long-term storage and refrigerated as working stock.
Availability and testing status
Being specific, because it differs:
- Retatrutide is currently stocked, and lot KP-RETA-2602 carries an
- independent certificate from Krause Analytical reporting 99.90% HPLC purity.
- Tirzepatide has no analysed lot at present. There is no purity figure
- we can quote for it, and we will not borrow one from another compound or a
- previous supplier.
