Melanotan II

Melanotan II is a synthetic cyclic analogue of alpha-melanocyte-stimulating hormone. The catalogue records the structure as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2: an acetylated norleucine residue followed by a lactam ring closed between aspartate and lysine, with a D-phenylalanine in place of the natural L form. Both the ring and the D-amino acid are described in the literature as conformational constraints that resist enzymatic breakdown relative to the linear hormone.

CAS 121062-08-6, formula C50H69N15O9, molar mass 1024.18 g/mol. It is often abbreviated MT-2.

Melanotan II appears in melanocortin receptor pharmacology as a non-selective agonist, with reported activity across the MC1, MC3, MC4 and MC5 receptors, which is what makes it a common reference compound in that literature.

Published studies have reported melanogenesis in pigment cell cultures through MC1 receptor activation, and have used the compound to probe MC4 receptor signalling in rodent models. It is frequently used as the non-selective comparator when a receptor-selective analogue is characterised.
